Targeting the cytokine storm: new therapies in the treatment of paediatric haemophagocytic lymphohistiocytosis. (PubMed, Expert Rev Clin Immunol)
Emapalumab, alemtuzumab and ruxolitinib show promising results, however widespread use is limited by small, non-randomized trials in a heterogenous population. Further international collaborative efforts to develop clinical trials to put conventional chemotherapy treatments head-to-head with these newer therapies is critical for advancement of these agents.

CD48 expression in T-lymphoblastic leukemia/lymphoma and mature T-cell neoplasms: role in detecting measurable residual disease and potential confounders, a single-institution retrospective review. (PubMed, Am J Clin Pathol)
CD48 expression is diminished compared with normal background T cells in most cases of T-ALL at diagnosis and posttherapy, whereas it was typically retained in mature T-cell/natural killer-cell neoplasms. While CD48 levels may fluctuate after chemotherapy in T-ALL, reduced CD48 can be used as a marker of immaturity in most MRD cases. Potential confounders in the assessment of CD48 include alemtuzumab therapy and the presence of GPI-deficient populations.

First-in-Human Study of IL15-Activated Cytokine-Induced Killer Cells After Allogeneic HCT Shows Durable Remission and Serotherapy-Associated Immune Reconstitution in Leukemia. (PubMed, J Clin Oncol)
IL15-CIK monotherapy is feasible and safe and demonstrates promising relapse-preventive activity after hematopoietic stem-cell transplantation. Clinical outcomes are strongly influenced by disease burden at treatment initiation and previous serotherapy, supporting optimized patient selection and timing in future post-transplant immunotherapeutic strategies.

A Unique Case of Concomitant T-Prolymphocytic Leukemia and B-Cell Acute Lymphoblastic Leukemia. (PubMed, EJHaem)
One proposed mechanism for the development of both hematologic malignancies in this patient is the acquisition of a KMT2A rearrangement, raising the possibility of clonal evolution resulting in therapy-related or secondary leukemia. Another explanation is the presence of a common clonal stem cell progenitor harboring a JAK3 mutation.
