Uterine Myxoid Inflammatory Myofibroblastic Sarcoma (MIMS) Harboring Pathogenic KRAS Mutations and Expressing ER, PR, and CD10. (PubMed, Int J Gynecol Pathol)
No gene fusions in PDGFRA/B, JAK1, PML, ALK, ROS1, PLAG1, or any other gene were identified by whole transcriptomic RNA sequencing. Given its deceptively bland appearance and its propensity, at least in the uterus, to express both CD10 and ER with only focal SMA expression, MIMS can be mistaken for an endometrial stromal tumor, an ALK and ROS1-negative uterine inflammatory myofibroblastic tumor, or other cytologically bland fusion-driven uterine mesenchymal neoplasms.

Particulate Hexavalent Chromium Inhibits RAD51 Paralogs Necessary for RAD51 Filament Formation and Stabilization During Homologous Recombination Repair. (PubMed, Occup Health (Auckl))
Cr(VI) also inhibited overall RAD51D protein expression, as well as its interaction with RAD51. These findings suggest that Cr(VI) inhibits all RAD51 paralogs, but RAD51D might be an early target of Cr(VI), leading to the loss of RAD51 filament formation and function and the overall inhibition of homologous recombination repair.

We observe durable PARPi effectiveness for patients with OVCA and HRR gene homozygous loss in routine practice. We demonstrate the importance of detecting and reporting homozygous loss in HRR genes, an aspect not standard to all sequencing assays.

Cancer mutations in RAD51 and its paralogues. (PubMed, PLoS One)
A comparison of mutations reported on COSMIC with those reported on ClinVar revealed that many mutations primarily in RAD51C and RAD51D are also hereditary. Thus, it appears that an apparent low level of RAD51 mutations in cancer cells is compensated by an increase in paralogues mutations.

Whole exome sequencing reveals rare DNA repair gene variants in BRCA1/2-negative Arab early-onset breast cancer patients. (PubMed, Sci Rep)
Several patients carried multiple deleterious variants, suggesting a possible polygenic contribution to EOBC predisposition. These findings expand the spectrum of rare DNA repair gene variants observed in BRCA1/2-negative early-onset breast cancer and provide candidate variants for further investigation, rather than establishing definitive causal associations.
