    Integrating breast tumour homologous recombination deficiency status to aid germline BRCA1 and BRCA2 variant classification. (PubMed, EBioMedicine)
    Analysis across multiple tumour whole-genome sequencing datasets has shown that HR status prediction algorithms can separate profiles for BRCA1 and BRCA2 pathogenic variants and provide further evidence at increased weight to aid in the classification of germline BRCA1 and BRCA2 variants. Tumour sequencing offers a promising strategy for reducing the uncertainty in germline variant interpretation.

    BCDX2-CX3 and DX2-CX3 complexes assemble and stabilize RAD51 filaments. (PubMed, Nature)
    These distinct assemblies are differentially regulated by ATPase activity, defining a dynamic BCDX2-CX3 "loader" and a stable DX2-CX3 "anchor" that provide functional modularity to the HR machinery. This work provides a unifying mechanism for human RAD51 paralog function and delivers an atomic blueprint for interpreting disease-causing mutations.

    RAD51C-XRCC3 complex regulates FANCM-mediated R-loop resolution to safeguard genome integrity. (PubMed, Sci Adv)
    The CX3 complex-mediated R-loop resolution is independent of its fork maintenance function. Collectively, we demonstrate a previously unidentified role of the CX3 complex in preventing R-loop-induced genome instability by regulating FANCM-mediated R-loop resolution.
