Case Report: The revelation of a new pathogenic variant in the POT1 gene in a patient with a pediatric high-grade glioma and a renal cell carcinoma. (PubMed, Front Oncol)
This is the first reported case of a young adult carrying a new pathogenic POT1 variant who developed a pediatric high-grade glioma and a renal tumor in early adulthood. This report expands the clinical spectrum of POT1-associated tumors with early-onset and underscores the relevance of genetic testing for patient management and family counseling.

Rapid On-Site Next-Generation Sequencing: An Alternative to Single-Gene and Send-Out Testing in Non-Small Cell Lung Cancer and Colorectal Cancer in a Community Pathology Laboratory Setting. (PubMed, J Mol Diagn)
In CRC, TAT was 4.1, 5.3, and 10.2; QNS 0%, 0%, 7.5%; detection 63.9%, 62.5%, and 57.1%. OPA provides faster TAT and lower QNS rates with comparable detection of actionable alterations, supporting its use for community-based molecular testing in NSCLC and CRC.

Both genotype and DNA methylation subclassification have been integrated into clinical decision-making, incorporating pretransplant azacitidine, watch-and-wait approaches for favorable-risk patients, and emerging targeted therapies including MEK inhibitors. This review synthesizes recent advances in understanding JMML as a bona fide RASopathy; provides a diagnostic algorithm, molecular landscapes, and prognostic models; and highlights opportunities for molecularly targeted therapeutic intervention.

Comprehensive characterization of non-small cell lung cancer of different PD-L1 expression classes: a study of 1,038 Chinese patients. (PubMed, J Thorac Dis)
This study enriches the current volume of evidence on histopathologic, genetic, immunologic correlates of PD-L1 expression and, to our knowledge, is the first comprehensive analysis of arm-level alterations. Further studies are warranted to validate these finding and to determine their associations with clinical outcomes.

Response to crizotinib treatment for ROS1 p.H1999N mutation and secondary EGFR p.V774M mutation after drug resistance: a Case Report. (PubMed, Front Pharmacol)
Subsequently, the patient received furmonertinib targeted therapy. Herein, we discuss the diagnostic challenges and the potential pathogenic mechanisms of this novel mutation. How to identify rare genes and translate their identification into clinical benefits is a worthwhile avenue for exploration in our future work.
