In addition, tumor cells exhibited a type II EBV latency pattern (LMP1+/EBNA2-) in 84.6% of cases, and LMP1 expression correlated with larger tumor size (p=0.021). Taken together, these findings support a role for EBV-driven activation of the cyclin D1 pathway and suggest that EBV-LMP1-CCND1 signaling may contribute to the pathogenesis of IFDCS.

This single-arm phase II trial enrolled patients with EPSCC to investigate the activity of first-line durvalumab (1500 mg every 3 wk) and 4 to 6 cycles of chemotherapy (either cisplatin or carboplatin) with etoposide, followed by maintenance durvalumab 1500 mg every 4 weeks until disease progression. This study reveals that durvalumab plus chemotherapy is safe and has some activity in rare small cell cancers outside the lung, but benefits are modest and short lived. In contrast, GO analysis demonstrated selective enrichment in PM tumors for molecular functions related to ATP binding, nucleotide binding, and protein kinase activity, consistent with enhanced bioenergetic demand and signaling intensity. These findings support refined biological stratification of mCC and the exploration of personalized, metastasis-directed strategies, potentially incorporating immunological modulation in OM disease.

In metastatic colon cancer, the presence of genome-wide copy number gain may delineate a tumor subset with distinctive clinicopathological and molecular characteristics. Further studies are warranted to elucidate the biological significance of these features and to explore their potential implications for tumor evolution, treatment response, and clinical stratification.
